Life-cycle-informed interpretation of microalgal bioproduct production and image-based cell-state analysis

Abstract


Microalgae produce a variety of high-value bioproducts, such as neutral lipids, polyunsaturated fatty acids, fucoxanthin, astaxanthin, and other pigments. Production studies typically quantify biomass as dry mass or estimate growth based on cell density, alongside biochemical metrics like fatty acid profiles and pigment contents. However, these bulk measurements do not reveal which coexisting life-cycle stages or physiological states were responsible for observed changes. This review offers a structured synthesis of evidence related to life-cycle stages, physiological states, and population-level events that impact target compound allocation and overall process performance. The evidence was analyzed based on perturbation, observable cell state, image-derived feature groups, biochemical endpoints, process outcomes, and validation strength. Specific emphasis was placed on Haematococcus, Chlamydomonas, and Tisochrysis/Isochrysis, as these taxa have available stage-resolved evidence or validation from single-cell to bulk assessments. Cross-study comparisons indicate that similar bulk biochemical responses can emerge from different population state compositions, while seemingly similar phenotypes can yield varying process outcomes. For example, a self-settling state of Tisochrysis combined harvestability with productivity in fucoxanthin and docosahexaenoic acid, whereas non-motile aggregated states of Haematococcus indicated stress adaptation, storage allocation, and trade-offs with rapid proliferation. These differing outcomes are attributed to factors such as state origin, reversibility, growth costs, target compound allocation, and harvest benefits, rather than aggregation alone. Image-derived variables were categorized into morphological, fluorescence, and population-event indicators. While these indicators facilitate state tracking, they do not correspond directly to biological states and require validation through methods such as time-series microscopy, gas chromatography or lipidomics, high-performance liquid chromatography or mass spectrometry, pulse-amplitude-modulated fluorometry, and independent cell counts. This evidence-based framework connects cell state interpretation to strain screening, stress diagnosis, harvest timing, scale-up assessment, and bioproduct quality control.



미세조류 유용물질 생산의 생활사적 해석과 영상 기반 세포상태 분석

초록


    1. 서 론

    미세조류는 빠른 생장과 높은 광합성 효율을 바탕으로 바이오매스, 식품·사료, 바이오연료 및 기능성 소재 생산에 활용된다(Spolaore et al. 2006;Brennan and Owende 2010;Mata et al. 2010). 주요 유용물질은 저장지질인 트리아실글리세롤(triacylglycerol, TAG), 다중불포화지방산(polyunsaturated fatty acids, PUFAs)인 알파-리놀렌산(α-linolenic acid, ALA), 에이코사펜타엔산(eicosapentaenoic acid, EPA)과 도코사헥사엔산(docosahexaenoic acid, DHA) 그리고 카로티노이드계 색소인 푸코잔틴과 아스타잔틴으로 구분할 수 있다(Guschina and Harwood 2006;Hu et al. 2008;Adarme-Vega et al. 2012;Maltsev and Maltseva 2021). 국내에서도 담수 미세조류 Tetradesmus obliquus의 배양 조건을 단계적으로 최적화하여 바이오매스와 지방산 함량을 높인 연구가 보고되어, 환경 조건과 생산 지표의 연결이 중요한 연구축임을 보여준다(Yun et al. 2025).

    기존 연구는 건조중량, 세포밀도, 지방산 조성, 색소 함량과 같은 배양액 전체의 평균값을 중심으로 생산성을 평가해 왔다. 그러나 시료 전체에서 측정한 값은 서로 다른 세포상태의 비율과 상태별 단위세포당 함량이 합쳐진 결과이다. 예를 들어, 같은 TAG 또는 아스타잔틴 함량이라도 모든 세포에서 함량이 완만하게 증가한 경우와 일부 저장물질 축적 세포가 증가한 경우는 생장 지속성, 수확 시점 및 유용물질의 균질성 측면에서 의미가 다르다. 녹조류 Haematococcus pluvialis에서는 운동성 녹색 세포에서 비운동성·palmelloid·적색 휴면상태로의 전환이 아스타잔틴, 지질체 및 세포벽 변화와 연결되고(Kobayashi et al. 1993;Boussiba 2000;Wayama et al. 2013;Shah et al. 2016;Fang et al. 2020), 녹조류 Chlamydomonas reinhardtii에서는 질소상태가 세포주기 정지, 휴지상태, TAG 축적 및 회복기 막지질 재구성을 조절한다(Spudich and Sager 1980;Cross and Umen 2015;Takeuchi and Benning 2019;Jang et al. 2020;Young and Shachar-Hill 2021). 따라서 생산 지표의 변화는 생활사 단계와 생리상태의 구성 변화로 분해하여 해석할 필요가 있다.

    이러한 분해에는 개별 세포의 형태와 형광을 반복 측정하는 영상 기반 단일세포 계측이 유용하다. 이 접근의 목적은 장비 자체를 소개하는 것이 아니라, 배양액 전체에서 측정된 생화학값을 구성하는 세포군의 상태 분포를 관측하는 데 있다. 환경생물학에서도 형태·분자계통·독성 자료를 결합한 유해 와편모조류 평가(Moon et al. 2024), 형태·생태·분자 자료를 통합한 남세균의 다상적 동정(Kim et al. 2025), 신속 영상·광학 지표와 기준 측정 및 독소 확인을 목적별로 분리한 유해 남세균 대발생 의사결정 체계가 제시되었다(Moon et al. 2026). 이들 연구는 신속한 관측 지표도 독립적인 기준법과 연결되어야 생물학적·공정적 판단에 활용할 수 있음을 공통적으로 보여준다. 영상 유세포 분석과 온라인 흐름 현미경 역시 형태·형광 정보를 세포 단위로 제공하지만(Havlik et al. 2013;Hyka et al. 2013;Hildebrand et al. 2016;Haberkorn et al. 2021;Kraus et al. 2022), 유용물질 함량과 조성에 관한 결론은 지방산·색소·광합성·바이오매스 자료와의 검증을 거쳐야 한다.

    본 총설의 목적은 미세조류 유용물질 생산 문헌을 생활사 단계, 생리상태 및 집단사건이라는 공통 비교축으로 재배열하고, 영상 유래 지표가 어떤 조건에서 이들 상태를 구분하는 데 기여하는지 평가하는 데 있다. H. pluvialis, C. reinhardtii 및 Tisochrysis/Isochrysis 계열을 중점 사례로 선택한 이유는 생활사 또는 성장 단계와 유용물질 축적의 연결이 보고되어 있고, 일부 연구에서 단일세포 신호와 생화학 정량 또는 공정 성능의 연계가 검증되었기 때문이다. 와편모조류와 기타 미세조류 사례는 생활사 단계-대사산물 연결의 적용 범위와 한계를 비교하기 위해 사용하였다.

    2. 문헌 범위와 비교 종합 방법

    본 총설은 관련 연구를 개별 사례의 연속으로 제시하지 않고 비교 가능한 항목으로 재배열한 구조화된 비정량 문헌 종합(structured qualitative evidence synthesis)을 사용하였다. 핵심 문헌은 2026년 7월까지 공개된 동료심사 문헌 중(1) 생활사·성장 단계 또는 세포주기와 유용물질·공정 결과를 직접 연결한 연구, (2) 단일세포 형태·형광 신호를 시료 전체의 생화학값과 보정한 연구, 또는(3) 응집·침강·휴지·휴면상태를 성장성·수확성·대사산물과 함께 평가한 연구로 선정하였다. 산업적 활용에 관한 일반 총설과 장비 중심 연구는 배경 또는 분석 한계를 설명하는 보조 문헌으로 구분하였다.

    관련 문헌은 2026년 7월까지 공개된 동료심사 논문을 대상으로 microalgae와 life cycle, cell state, growth phase, cell cycle, imaging flow cytometry, single-cell analysis, lipid accumulation, fatty acid, pigment accumulation, aggregation, settling 및 harvestability를 조합한 표적 검색으로 탐색하였다. 제목·초록과 전문을 단계적으로 검토하여 위의 포함 기준에 부합하는 연구를 선별하고, 핵심 문헌의 참고문헌을 추가로 검토하였다. 서지정보와 DOI는 Crossref와 출판사 원문 페이지에서 확인하였다.

    각 핵심 문헌에서 분류군, 배양 또는 환경 요인, 관찰된 생활사 단계, 생리상태, 집단사건, 영상 특징, 생화학 지표, 공정 결과 및 독립 검증법을 추출하였다. 이후 증거를(i) 단계-유용물질 연결, (ii) 단일세포-시료 전체 보정, (iii) 상태-공정 특성 연결, (iv) 자동분류와 외부 검증의 네 문헌군으로 비교하였다. H. pluvialis, C. reinhardtii와 Tisochrysis/Isochrysis는 이 항목 가운데 둘 이상을 직접 연결한 연구가 축적되어 핵심 비교 대상으로 사용하였다. 기타 미세조류와 harmful algal bloom (HAB) 관련 사례는 프레임워크의 적용 범위, 환경생물학적 의미 및 일반화 한계를 평가하는 비교 자료로 사용하였다.

    분류군, 처리 조건, 영상장비 및 정량 지표의 이질성이 커서 효과크기를 통합하는 메타분석은 적용하지 않았다. 대신 연구 사이에서 결과 방향이 일치하는지, 같은 상태가 서로 다른 공정 결과를 보이는지, 차이가 종·성장 단계·상태 기원·가역성·검증법으로 설명되는지를 정성적으로 비교하였다. Table 1은 문헌군별 확립된 결론, 남은 불확실성 및 해석 전략을 제시하고, Table 2는 영상 지표와 후보상태 및 필수 검증법의 대응을 제시한다.

    3. 바이오매스·생화학 지표와 생활사 해석의 차이

    본 총설에서 바이오매스는 배양액 부피당 건조중량을 뜻하며, 바이오매스 생산성은 그 시간당 증가량으로 구분한다. 세포밀도는 이를 보완하는 개체수 지표이다. 유용물질 정량에는 GC-FID·GC-MS 또는 지질체학 기반 지방산·지질 분석과 HPLC-DAD·LC-MS 기반 색소 분석이 사용되고, 광합성 상태는 PAM 형광측정과 엽록소 분석으로 평가된다. 이들 기준법은 생산량과 유용물질 품질을 확인하는 핵심 수단이지만, 세포군 내부의 상태 분포를 직접 보여주지는 않는다. 국내 분리주 Eutreptiella sp.와 Picochlorum sp. 연구에서도 형태·계통·지방산 조성을 통합해 자원 가치를 평가했으나, 생산 지표와 생활사 단계의 정량적 연결은 후속 과제로 남아 있다(Kang et al. 2023, 2024).

    동일한 바이오매스 농도라도 생활사 단계, 생리상태 및 집단사건의 조성이 다를 수 있으므로 공정 성능과 유용물질 특성을 구분하여 해석해야 한다. 공정 특성에는 생장 지속성, 스트레스 내성, 배양 안정성, 수확성 및 배양 규모 확대 가능성이 포함되고, 유용물질 특성에는 저장지질, 지방산 및 색소의 단위 바이오매스당 함량, 조성 및 균질성이 포함된다. 활발히 분열하는 세포가 우점하는 배양과 지질·색소 축적, 비운동성, 휴지 또는 응집·침강 상태가 우점하는 배양은 동일한 바이오매스 농도를 보이더라도 유용물질 배분, 공정 확장성 및 적정 수확 시기가 다를 수 있다. 성장 단계가 지질군과 지방산 조성에 미치는 영향은 Isochrysis galbana와 T. lutea에서 반복적으로 보고되었으며(Fidalgo et al. 1998;Lin et al. 2007;da Costa et al. 2017), 따라서 배양일수만 기록하기보다 성장 단계, 상태 비율 및 단위상태당 유용물질량을 함께 기록해야 한다. 이러한 비교 논리와 상태 의존적 결과를 Fig. 1과 Table 1에 정리하였다.

    4. 생활사 및 세포상태 판독을 위한 관측 지표와 검증 원리

    영상 유래 지표는 기능에 따라 형태 지표, 형광 지표 및 집단사건 지표로 구분할 수 있다. 형태 지표에는 세포면적·직경·부피, 종횡비, 원형도, 경계와 질감이 포함되며 분열, 딸세포 쌍, 세포 확대 및 세포벽 변화의 후보 신호가 된다. 형광 지표에는 엽록소 자가형광, 지질 염색 신호 및 색소 관련 스펙트럼이 포함되며 광합성 상태와 저장물질·색소 축적의 후보 신호로 사용된다. 집단사건 지표에는 단일세포·다세포 이벤트 비율, 응집체 크기, 운동성 및 침강성이 포함된다. 이 세 범주는 관측값의 종류를 구분하는 것이며, 하나의 지표가 하나의 생물학적 상태에 고정적으로 대응한다는 뜻은 아니다.

    문헌 간 비교에서 단일세포 신호의 해석력은 독립적인 정량값과 연결될 때 높아졌다. T. lutea에서는 단일세포 형광과 푸코잔틴·지질의 대응이 제시되었고(Gao et al. 2020b), Euglena gracilis에서는 영상 유세포 분석으로 산출한 세포별 지질 생산성 지표가 시료 전체 오일 추출값과 상관을 보였다(Muñoz et al. 2018). Acutodesmus obliquus에서는 엽록소와 Nile Red 신호를 이용해 질소 결핍 중 지질 축적 하위집단을 구분했으며(Sandmann et al. 2018), H. pluvialis에서는 다중스펙트럼 영상 유세포 분석을 색소·엽록소 분포와 공정 단계의 추적에 적용하였다(Kraus et al. 2022). 또한 비표지 굴절률 단층촬영은 개별 세포의 지질 함량을 정량했고(Jung et al. 2018), 유세포 선별과 미세유체 홀로그래피는 각각 푸코잔틴 생산 세포의 선별과 배양 중 세포상태 추적에 적용되었다(Fan et al. 2021;Song et al. 2022). 공통점은 상태 이질성을 세포 단위로 확인했다는 점이고, 차이점은 형광의 대상, 종의 세포벽·자가형광, 사용 장비 및 기준 정량법이 달랐다는 점이다.

    따라서 형태 또는 형광 지표의 절대 임계값을 종 사이에 그대로 이전할 수 없다. 지질 신호는 염색 투과성과 세포 크기의 영향을 받고, 색소 관련 신호는 색소 조성·광합성 상태·장비 대역의 영향을 받는다(Čertnerová and Galbraith 2021;Verwee et al. 2024). 상태 판독은 시간에 따른 지표 조합과 독립적인 현미경 관찰로 수행하며, 유용물질 함량과 조성은 GC·지질체학 또는 HPLC·LC-MS, 생리상태는 PAM과 생존성, 바이오매스는 건조중량과 독립 세포계수로 검증해야 한다. 이는 신속 지표와 기준 측정의 역할을 분리하는 최근 환경 모니터링 체계와도 일치한다(Moon et al. 2026).

    5. 생활사 기반 세포군 상태 구조의 정의와 범위

    5.1. 생활사 단계, 생리상태 및 집단사건의 구분

    ‘생활사 기반 세포군 상태 구조’는 서로 다른 수준의 범주를 한데 묶는 명칭이 아니라, 배양액에서 동시에 관찰되는 세 수준의 분포를 함께 기록하는 해석 단위이다. 첫째, 생활사 단계는 영양세포, 분열세포, 딸세포 쌍 및 휴면 단계처럼 발생학적 순서나 세포주기 위치가 비교적 명확한 범주이다. 둘째, 생리상태는 지질 축적, 색소 축적, 휴지 또는 스트레스 적응처럼 기능과 대사 배분으로 정의되는 범주이다. 셋째, 집단사건은 응집, 운동성 소실, 자가침강 또는 불완전한 세포 분리처럼 둘 이상의 세포나 배양액의 물리적 거동으로 관찰되는 현상이다. 한 세포가 비운동성·색소 축적·응집상태를 동시에 가질 수 있으므로 이 범주들은 상호 배타적이지 않다.

    이 구분은 종 사이의 공통점과 차이점을 드러낸다. 녹조류 H. pluvialis에서는 운동성 소실, 세포 확대, 세포벽 강화, 지질체와 아스타잔틴 축적이 연속적인 생활사 전환 안에서 결합된다(Boussiba 2000;Wayama et al. 2013;Tang et al. 2024). C. reinhardtii에서는 질소 결핍에 따른 세포주기 정지와 휴지상태가 TAG 저장 및 재급식 후 막지질 회복과 연결되어 상태의 가역성이 강조된다(Takeuchi and Benning 2019;Young and Shachar-Hill 2021). 두 계열 모두 분열 감소와 저장물질 배분이 연결되지만, H. pluvialis에서는 세포벽·운동성·색소 변화가 강하고 Chlamydomonas에서는 영양상태에 따른 저장과 회복의 전환이 더 직접적으로 검증되었다.

    5.2. 다중 지표 조합을 통한 상태 관측과 문헌 간 비교

    상태 판독은 단일 지표보다 사전에 정의한 지표 조합과 시간적 궤적을 사용해야 한다. 예를 들어 세포면적 증가와 원형도 변화만으로 휴면상태를 확정할 수 없지만, 운동성 소실, 세포벽 또는 질감 변화, 엽록소·색소 신호 변화와 함께 지속될 경우 H. pluvialis의 비운동성 또는 휴면 관련 상태 후보로 분류할 수 있다. 분열세포와 딸세포 쌍은 세포 크기, 경계의 함몰, 연결된 세포 수 및 시간에 따른 분리 여부를 함께 사용한다. 지질 축적 상태는 지질 염색 신호, 세포부피 또는 지질체 면적을 조합하되 GC 또는 지질체학 정량으로 확인한다. 이와 같은 대응 관계와 필요한 검증법은 Table 2에 정리하였다.

    단일세포 지표와 시료 전체 정량값을 연결한 연구들은 서로 다른 검증 단계를 다루었다. Gao et al. (2020b)과 Muñoz et al. (2018)은 단일세포 신호가 유용물질 정량을 위한 선별 지표가 될 수 있음을 보였고, Sandmann et al. (2018)은 평균값 뒤에 숨은 지질 축적 하위집단을 분리하였다. Kraus et al. (2022)은 색소·형태 분포를 공정 단계와 연결하였다. 따라서 이 연구들의 공통 결론은 영상 신호가 생화학 분석을 대체한다는 것이 아니라, 같은 평균값을 만드는 서로 다른 세포군 구성을 구별한다는 데 있다. 반면 염색 기반, 자가형광 기반 및 비표지 방식은 신호 생성 원리가 다르므로 임계값과 정확도를 직접 비교해서는 안 된다.

    생활사 단계와 대사산물의 연결은 유용물질 생산 이외의 환경생물학적 사례에서도 확인된다. Crypthecodinium cohnii에서는 배양 중 형태와 저장물질 변화가 실시간으로 추적되었고(Marbà-Ardébol et al. 2017), Alexandrium pacificum에서는 영양세포에서 접합자·휴면포자로의 전환이 패류독소 함량과 조성 변화에 연결되었다(Jiang et al. 2025). Centrodinium punctatum에서도 발달 단계별 형태·형광과 독성 조성이 함께 변하였다(Jiang et al. 2026). 이 사례들은 특정 독성 결과를 유용물질 생산으로 일반화하기 위한 것이 아니라, 세포밀도만으로 설명되지 않는 단계별 대사 배분을 검증한 비교 근거로 사용하였다.

    5.3. 영상 기반 상태 분류의 역할과 적용 범위

    영상 기반 세포상태 분석의 역할은 시간대별 시료에서 형태·형광·집단사건 특징을 추출하여 반복 가능한 후보상태 비율과 전환속도로 변환하는 데 있다. 구현 방식은 규칙 기반 게이팅, 계산된 특징을 이용한 기계학습 및 원영상을 이용한 딥러닝으로 구분되며, 각 방법의 전제와 검증 요건은 Section 7에서 다룬다. 어느 방식에서도 관측 특징과 생물학적 상태는 일대일 관계가 아니므로 자동분류 결과는 확정상태가 아닌 후보상태로 보고하고, 혼합상태와 불확실성을 유지해야 한다. 배양 시료 채취, 영상 획득, 세포·응집체 분할과 사건 품질관리(event quality control), 특징 추출, 자동 후보 세포상태 분류, 독립 검증 및 공정 해석의 6단계를 Fig. 2에 제시하였다.

    6. 생활사 기반 유용물질 생산 연구의 비교 종합

    6.1. 성장 단계·세포주기와 지방산·중성지질 축적

    스트레스 중심 모델은 질소 제한과 같은 처리가 탄소를 TAG로 재배분한다는 평균적 반응을 설명하지만, 생활사 기반 해석은 그 반응이 어느 성장 단계와 세포군에서 발생하고 회복 가능한지를 묻는다. Isochrysis/Tisochrysis에서는 성장 단계가 지방산 조성, 중성지질 및 저장지질에 큰 영향을 미쳤고(Fidalgo et al. 1998;Huerlimann et al. 2014;da Costa et al. 2017), C. reinhardtii에서는 질소 결핍 중 세포주기 정지와 TAG 저장이 결합되고 재급식 후 저장 지방산이 막지질로 되돌아갔다(Takeuchi and Benning 2019;Young and Shachar-Hill 2021). 따라서 TAG 증가는 고정된 최종 상태가 아니라 증식-저장-회복 사이의 시간 의존적 전환이다.

    실제 생산성 평가에서는 지질 염색 강도 하나보다 지질 축적 세포의 비율, 세포당 지질 신호, 세포부피, 분열세포·딸세포 쌍 비율 및 성장률을 함께 사용해야 한다. 세포당 신호와 지질 축적 세포 비율이 증가하면서 건조중량 또는 세포수 기반 생산성이 유지되고 GC 기반 total fatty acids (TFA)·TAG가 증가하면 생산적 배분으로 해석할 수 있다. 반대로 신호 증가가 분열 정지, maximum quantum yield of photosystem II (Fv/Fm) 저하, 생존성 감소 및 낮은 바이오매스 생산성과 동반되면 생산적 축적보다 스트레스 과부하를 반영할 가능성이 높다. 이 구분은 기존 스트레스 모델에 세포군 구성과 회복 가능성을 추가한다.

    6.2. Tisochrysis/Isochrysis: 생산성·자가침강·세포군 구조의 통합

    Tisochrysis/Isochrysis 문헌은 시간 순서보다 기능에 따라 네 연구군으로 종합할 수 있다. 첫째, 성장 단계와 질소상태 연구는 DHA, PUFAs, 중성지질 및 알케논 배분이 배양 단계에 의존함을 보여준다(Fidalgo et al. 1998;Huerlimann et al. 2014;da Costa et al. 2017). 둘째, 푸코잔틴 공정 최적화와 단일세포 형광 연구는 시료 전체 생산량과 세포별 색소·지질 신호를 연결하였다(Gao et al. 2020a, 2020b). 셋째, 선별된 자가침강 상태와 광질 유도 연구는 세포군 특성 자체가 수확성과 푸코잔틴·DHA 생산성의 선택 대상이 될 수 있음을 보였다(Gao et al. 2021a, 2021b). 넷째, 최근 배양 연구는 광질과 식물호르몬 조건에서 생장, Fv/Fm, 푸코잔틴, 지방산 및 단일세포·응집 지표를 함께 평가하였다(Kang et al. 2026). 이 문헌군의 공통 결론은 높은 함량만으로 최적 조건을 정할 수 없고 성장 단계, 세포별 배분, 응집 안정성 및 수확성을 함께 평가해야 한다는 것이다. 다만 형광 신호-유용물질량의 보정 관계와 자가침강의 이점은 균주·장비·배양 규모에 의존하므로 다른 종에 직접 이전할 수 없다.

    6.3. Haematococcus: 생활사 전환과 아스타잔틴 생산의 상태 의존성

    H. pluvialis의 기존 생산성 중심 해석은 스트레스 조건과 최종 아스타잔틴 함량의 관계를 강조한다. 생활사 기반 해석은 같은 함량 증가를 운동성 녹색세포의 광범위한 변화, 비운동성 세포의 증가 또는 적색 휴면상태의 축적으로 구분한다. 이 전환에는 분열률 감소, 세포 확대, 세포벽 강화, 지질체 증가, 엽록체 재편 및 아스타잔틴 축적이 서로 다른 시간척도로 결합된다(Boussiba and Vonshak 1991;Boussiba 2000;Collins et al. 2011;Wayama et al. 2013;Tang et al. 2024). 따라서 세포 크기·원형도·운동성, 엽록소 및 색소 관련 신호, 지질체 또는 지질 염색 신호, 응집체 비율을 시간에 따라 측정하고 HPLC 아스타잔틴, PAM, 생존성 및 독립 세포계수와 연결해야 한다. 모든 세포의 색소 신호가 증가하면서 생존성과 생산성이 유지되는 경우와 비운동성·응집세포가 우점하면서 함량이 증가하는 경우는 유도 시점, 수확창 및 후속 배양 가능성이 다르다.

    6.4. 영양염 스트레스: 축적 촉진과 공정 실패의 분기

    영양염 스트레스는 세포분열보다 탄소 동화가 상대적으로 유지되는 범위에서는 남는 환원력과 탄소를 TAG 또는 저장물질로 전환할 수 있다. 그러나 제한이 심해 광합성 전자전달, 단백질 합성, 막 유지 및 생존성이 함께 저하되면 탄소 공급 자체가 감소하여 유용물질 축적과 바이오매스 생산이 모두 감소할 수 있다. 따라서 ‘질소 스트레스’는 단일 처리명이 아니라 영양염 농도, 노출시간, 성장 단계 및 회복 가능성에 따라 생산적 재배분과 스트레스 과부하로 갈라지는 연속체이다(Miller et al. 2010;White et al. 2011;Takeuchi and Benning 2019;Young and Shachar-Hill 2021).

    이 분기점은 앞서 제시한 C. reinhardtii의 가역적 TAG 저장과 담수성 Desmid인 Cosmarium tinctum의 질소 과잉에서 나타난 Fv/Fm 저하, 딸세포 쌍 축적 및 TFA 축적 억제의 대비에서 드러난다(Takeuchi and Benning 2019;Kang et al. 2025). T. obliquus의 단계적 배양에서는 먼저 바이오매스를 확보한 뒤 질소·인 결핍을 적용하여 지방산 함량을 높였다(Yun et al. 2025). 세 사례의 공통 전략은 바이오매스 확보 단계와 스트레스 유도·수확 시점을 구분하는 것이다. 이러한 단계 분리 전략은 생장과 지질 생산성의 균형을 강조한 2단계 배양 연구와 일치한다(Griffiths and Harrison 2009;Rodolfi et al. 2009;Aziz et al. 2020). 분열세포·딸세포 쌍 비율, 지질 신호, Fv/Fm, 건조중량 및 지방산을 함께 추적하면 생산적 유도 구간과 공정 실패 구간을 구분할 수 있다.

    6.5. 응집, 자가침강 및 EPS: 수확 이점과 스트레스 비용의 조건부 균형

    응집, 침강 및 extracellular polymeric substances (EPS) 형성은 구분하여 측정해야 한다. 응집은 여러 세포가 하나의 집단을 이루는 상태이고, 침강은 그 집단 또는 세포가 중력에 의해 분리되는 공정 특성이며, EPS는 부착·응집·점도·추출성에 영향을 주는 세포외 물질이다(Li et al. 2021;Zhou et al. 2024). 세 현상은 함께 나타날 수 있지만 동일하지 않다. 수확성의 이점은 침강속도와 회수율로 평가하고, 생물학적 비용은 성장률, 생존성, 광합성, 상태의 가역성 및 유용물질 조성으로 평가해야 한다.

    Gao et al. (2021a)과 Tang et al. (2024)의 결과는 표면적으로 상반되지만, 응집 자체보다 상태가 형성된 경로가 다르다. Gao et al. (2021a)의 T. lutea는 fluorescence-activated cell sorting (FACS)로 선별된 무편모·자가침강 상태로, 파일럿 규모에서 수확성과 함께 바이오매스, 푸코잔틴 및 DHA 생산성이 유지 또는 향상되었다. Tang et al. (2024)의 H. pluvialis 비운동성 세포군은 가역적 bet-hedging의 일부로서 세포 확대, 두꺼운 세포벽, 응집, 항산화 및 저장물 축적을 보였지만 빠른 증식 상태와는 대조적이었다. 따라서 자가침강의 생산적 가치는(1) 선발된 형질인지 스트레스 유도상태인지, (2) 재분산·재성장이 가능한지, (3) 성장 손실보다 수확 이득이 큰지, (4) 목표물질의 단위부피당 생산성이 유지되는지, (5) 유용물질 품질과 추출성이 안정적인지에 따라 결정된다. 공정 전략은 응집을 일률적으로 촉진하거나 억제하는 것이 아니라 이 다섯 항목을 배양 단계별로 평가하는 것이다.

    7. 영상 기반 자동분류의 방법론과 발전 방향

    자동분류의 방법은 규칙 기반, 특징 기반 기계학습 및 딥러닝으로 나뉜다. 규칙 기반 분류는 크기·형광·형태의 사전 임계값으로 세포상태를 구분하므로 해석이 쉽다. 특징 기반 기계학습은 원형도, 질감, 자가형광과 같이 계산된 변수를 이용해 분류기를 학습한다. 합성곱신경망(convolutional neural network, CNN)은 이미지의 국소 패턴을 계층적으로 학습하는 딥러닝 모델로, 복잡한 세포 형태를 직접 분류할 수 있다. Işıl et al. (2021)은 비표지 영상 유세포 분석과 CNN을 결합해 처리 조건에 따른 미세조류 집단 조성과 형태 분포를 구분하였다. 관련 방법론은 실시간 딥러닝 유세포 분석(Li et al. 2019), 불균형 플랑크톤 영상 자료의 분류(Kerr et al. 2020), 라벨 없는 분류와 이상탐지(Pastore et al. 2020)로 확장되어 왔다. 다만 모델의 목적은 대량 이미지에서 세포상태를 반복적으로 추적하는 것이며, 유용물질량을 직접 정량하는 것은 아니다.

    자동분류 결과는 ‘특징 조합-후보상태-독립 검증-공정 해석’의 순서로 사용해야 한다. 예를 들어 큰 입자, 운동성 소실 및 높은 색소 신호가 함께 나타나면 비운동성 색소 축적 상태의 후보로 분류할 수 있지만, 성장 단계, 생존성 및 HPLC 색소 함량을 확인하기 전에는 생산적 상태로 확정할 수 없다. 학습자료의 종, 장비, 이미지 품질 및 상태 비율이 달라지면 성능이 저하될 수 있으므로(Maracani et al. 2023), 동일 배양계 내부 검증과 다른 배양회차·장비·균주를 이용한 외부 검증을 구분해야 한다. 불확실하거나 여러 상태가 겹치는 세포는 강제로 단일 범주에 배정하지 않고 혼합 또는 미분류 상태로 남기는 것이 적절하다.

    재현성과 일반화를 위해서는 이미지 획득 조건, 전처리, 상태 주석 기준, 상태별 이미지 수, 학습·내부 검증·외부 검증 자료의 분리, 불균형 처리, 모델 구조 및 성능 지표를 보고해야 한다. 정확도만으로는 드문 휴면상태나 응집체의 누락을 감추므로 정밀도, 재현율, F1 값과 혼동행렬을 함께 제시한다. 이러한 표준화는 같은 명칭의 상태가 연구마다 다른 특징으로 정의되는 것을 방지하고, 분류 성능이 새로운 배양 조건에서도 유지되는지를 평가하게 한다. 영상 지표, 후보상태, 생물학적 의미, 필수 검증 및 주요 모호성의 대응을 Table 2에 요약하였다.

    8. 공정 적용의 한계와 검증 전략

    공정 적용의 첫 번째 한계는 표본 단위이다. 영상 장비에서 응집체 하나는 하나의 이벤트로 기록될 수 있지만 여러 세포를 포함하므로 이벤트 수를 세포수로 직접 환산할 수 없다. 응집이 증가하는 배양에서는 독립 세포계수, 건조중량, 응집체 크기분포 및 침강속도를 함께 기록해야 한다.

    두 번째 한계는 종·장비·배양 조건 의존성이다. 동일한 형광 강도나 크기라도 세포벽, 자가형광, 염색 투과성 및 광환경이 다르면 의미가 달라지므로 임계값과 보정식은 조건별로 재검증해야 한다.

    세 번째 한계는 실험실에서 대량배양으로의 이전이다. 플라스크, 기포탑, 광생물반응기 및 야외 배양은 혼합, 전단력, 광주기, pH·CO2 및 영양염 구배가 다르며, 이 차이가 상태 비율과 응집에 영향을 미칠 수 있다. 규모 이전 검증은 같은 상태 정의를 적용한 뒤 상태 비율, 성장률, 유용물질 정량, 수확 회수율 및 배양 안정성을 동시에 비교해야 한다. 실험실 분류 정확도가 높더라도 공정 의사결정의 이득이 입증되지 않으면 품질관리 지표로 채택하기 어렵다.

    따라서 적용 단계는 탐색, 확인 및 의사결정으로 구분하는 것이 타당하다. 탐색 단계에서는 영상 지표로 후보 균주와 이상 상태를 빠르게 선별한다. 확인 단계에서는 기준 생화학·생리 분석으로 상태의 의미를 검증한다. 의사결정 단계에서는 상태 지표가 수확 시점, 스트레스 종료 또는 배양 중단을 기존 방식보다 빠르거나 안정적으로 판단하게 하는지 평가한다. 이 단계 구분은 영상 분석을 보조 해석 도구로 제한하는 것이 아니라, 각 증거가 뒷받침할 수 있는 결론의 범위를 명확히 하는 검증 전략이다.

    9. 결 론

    문헌 비교에서 일관되게 확인된 점은 유용물질 생산량이 배양액의 단일 평균 반응이 아니라 서로 다른 생활사 단계, 생리상태 및 집단사건의 조합으로 형성된다는 것이다. 성장 단계와 세포주기는 저장과 회복의 시간적 위치를 결정하고, 운동성 소실·응집·침강은 목표물질 축적과 수확성에 이득 또는 비용을 동시에 줄 수 있다. 특히 T. lutea의 자가침강 상태와 H. pluvialis의 비운동성 응집 상태는 비슷한 외형이 상태의 기원과 성장비용에 따라 다른 공정 결과를 낳는다는 점을 보여준다.

    따라서 앞으로의 연구에서는 평균 함량뿐 아니라 상태 비율, 단위상태당 유용물질량, 전환속도 및 회복 가능성을 함께 평가해야 한다. 영상 기반 단일세포 분석은 이 네 항목을 추적하는 데 유용하지만, 형태·형광·집단사건 지표를 독립적인 생리·생화학 자료와 연결할 때에만 생산성과 품질에 관한 결론을 지지할 수 있다. 이 증거체계는 균주 선별, 스트레스 종료, 수확 시점 및 규모 확대의 판단 체계를 생활사 기반으로 전환하는 검증 가능한 출발점을 제공한다.

    적 요

    미세조류의 유용물질 생산량은 배양계에 공존하는 여러 생활사 단계와 세포상태가 함께 만든 결과이므로 배양액 전체의 바이오매스·지방산·색소값만으로는 생산성과 공정 안정성을 충분히 설명하기 어렵다. 본 총설은 관련 문헌을 생활사 단계, 생리상태 및 집단사건으로 구분하고, 처리조건-세포상태-영상 지표-생화학 지표-공정 결과의 비교축으로 종합하였다. 문헌 비교 결과, 유사한 응집·비운동성 상태도 상태의 기원, 가역성, 성장비용 및 목표물질 배분에 따라 수확 이점 또는 스트레스 비용으로 작용하였다. 영상 지표는 형태, 형광 및 집단사건 지표로 구분할 수 있으나 생물학적 상태와 일대일로 대응하지 않으므로 시간적 관찰과 GC, HPLC, PAM, 건조중량 및 독립 세포계수를 이용한 검증이 필요하다. 이러한 증거 기반 해석은 균주 선별, 스트레스 진단, 수확 시점, 규모 확대 및 유용물질 품질관리의 판단 근거를 강화할 수 있다.

    Literature-derived evidence-synthesis framework linking cultivation drivers and environmental stressors to population-state composition and state-dependent outcomes in microalgal cultures. Microalgal cultures can contain coexisting life cycle stages, physiological states, and population-level events. As a result, cultures with the same biomass concentration may differ in state composition, leading to variations in bioproduct allocation, culture performance, and harvestability. Bioproduct outcomes include storage lipids, fatty acids (including DHA), and pigments. Arrows indicate conditional and potentially reversible relationships, where the direction and benefits depend on the intensity, duration, and interactions of cultivation drivers and stressors.     Operational workflow for image-based candidate cell-state classification, independent validation, and process interpretation. Time-resolved culture sampling preserves biological replicates and treatment metadata while capturing both single cells and aggregates. Bright-field, autofluorescence, lipid-staining, and pigment-spectral images undergo segmentation and quality control before features such as size, shape, texture, fluorescence, and aggregation are extracted. Automated models assign candidate life-cycle-related states instead of definitive biological states, with classification performance evaluated using precision, recall, F1 score, and confusion matrices.
